WATER TESTING INSTRUCTIONS
Sample/Test container Requirements
Use sterile, non-leaking bottles, typically provided by the lab, which contain sodium thiosulfate to neutralize chlorine residuals. If not, ensure the approved sterile container contains sodium thiosulfate for dechlorination of chlorinated samples.
- For SM 9223 B Colilert - Total Coliform and E. coli:
- Sterile, non-fluorescent borosilicate glass containers, OR sterile plastic containers that are non-leaching and made of materials such as High-Density Polyethylene (HDPE) or other autoclavable plastics.
Water sample bottles are available from Indepth Water Testing.
Sample Collection Guidelines
- Always collect cold water.
- Wear gloves during sample collection.
- Select a tap, remove any aerator or hose, and clean the faucet if necessary.
- Run cold water for 4–6 minutes (EPA recommendation), or until the temperature stabilizes, to ensure the sample represents the main line rather than the service line.
- Reduce water flow to a gentle stream to prevent splashing.
- Carefully open the sample container without touching the inside of the cap or bottle. Hold the cap by its outside edges and avoid setting it down.
- Fill the bottle to the 100 mL mark, ensuring there is air space . Do not overfill past this line.
- Cap the bottle tightly immediately after filling.

- Label the bottle immediately with the site, date, time of collection, and sampler name. Follow the lab’s chain of custody forms as required for analysis.
- Note the address of the property
- Date/time the sample was taken
- Specific sample location (i.e., kitchen sink, tub spout, yard hydrant)
- Your information (for results) including an email and phone number.
- Place sample bottles in a cooler with ice for transport. The EPA encourages holding samples below 10°C during transit, but do not allow samples to freeze.
- The sample should be kept cold (refrigerated or in a cooler). It must be delivered to the Indepth Water Testing lab within 24 hours of collection.
